Phenacetin Descrption
Phenacetin is a pain-relieving and fever-reducing drug, widely used from its introduction in 1887 until banned in the US by the FDA in 1983. Its use has declined because of its adverse effects, which include increased risk of certain cancers and kidney damage. It is metabolized into paracetamol, which replaced it in some over-the-counter medications following the ban on phenacetin.
Phenacetin Application
The antipyretic effect of phenacetin is stronger than the analgesic effect. The pharmacodynamic intensity is similar to that of aspirin, with slow and lasting action and low toxicity. The results showed that the product and its metabolite paracetamol had antipyretic effect. Because when phenacetin can not be converted into paracetamol by enzyme inhibitors, it still has obvious antipyretic effect. Therefore, the antipyretic effect after wire feeding is not only caused by paracetamol, its active product. The mild analgesic effect of phenacetin generally lasts for 3-4 hours, and the synergistic effect of phenacetin combined with salicylic acid enhances the analgesic effect. It is mainly used for antipyretic analgesia in small animals. This product is also one of the components of APC tablets.
Phenacetin is now being used as a cutting agent to adulterate cocaine in the UK and Canada, owing to the similar physical features of the two drugs.Due to low cost phenacetin is used for research into the physical and refractive properties of crystals. It is an ideal compound for this type of research.
Phenacetin Dosage
Analgesic mixtures containing phenacetin were previously marketed as tablets or capsules containing between 150 and 300 mg phenacetin. Common combinations were: 150 mg phenacetin, 230 mg aspirin, and 15 or 30 mg caffeine; or 150 mg phenacetin, 230 mg aspirin, 30 mg caffeine, and 8, 15, 30, or 60 mg codeine phosphate. Phenacetin alone was also available in 250 and 300 mg doses as tablets, and up to 500 mg doses as powder. The usual dose was 300 mg 4-6 times per day, and the daily dose was not to exceed 2 g.
